Different Methods for Detection and Diagnosis of Tomato Spotted Wilt Tospovirus Infection of some Cucurbit Host Plants and its Main Vector of Thrips Species in Field and Greenhouse in Egypt

The study recorded for the first time in Egypt presence of tomato spotted wilt virus (TSWV) on cucurbits, which was confirmed using several serological tests, like double antibody sandwich ELISA (DAS-ELISA); Dot-blot assay and Immuno capture polymerase chain reaction (IC-PCR).  By Dot blot immunoassay test against TSWV antisera, ability thrips species Thrips tabaci (Lind), palmi (Karny), Frankliniella occidentalis (​Pergande), transmission were asserted. In greenhouse, eleven plant determined as host range field samples showed similar symptoms. mechanical a positive bearing symptoms leaves Cucumber, zucchini, smoke, datura plants 15-20 days after inoculation with rates 46.5, 40, 46.5 86.5%, respectively.  DAS-ELISA antisera that greenhouse infected; otherwise, an amplification IC-PCR products 500 bp detected by agarose electrophoresis total RNA isolated from infected leaves.
